No, you infest the red ant queen in the base game, then befriend the other too, then you have an infected in ng+1, and you can also feed the fire ant queen again to max out the mutation. Then you can infect the black (in ng+1), and the fire in ng+2.

One thing you can do for raw science is go to the hedge and break the web sacks and if your lucky you can get up to 3 infused spiderlings it helped me get teleporters
